Title Characterizing hookah smoking-induced transcriptional regulations in the human bronchial epithelial cell

Summary Hookah smoking is becoming increasingly popular worldwide and recent epidemiological studies indicate that it is significantly associated with respiratory diseases, and could contain constituents more harmful than cigarette smoking. However, the gene expression alterations and mechanisms of disease development induced by the hookah constituents are still unclear. Here, we propose a new experimental design that compared the constituents of hookah smoking generated with different heating ways and w/wo tobacco, for studying the harmful effects on respiratory cells (30 samples among five conditions).
 
Overall design BEAS-2B cells were exposed to various conditions of hookah smoking, including internal control (IC), charcoal with no shisha (C), charcoal with non-tobacco (herbal) shisha (CNT), charcoal with tobacco shisha (CT), and electronic heater with tobacco shisha (ET) for 30 min while on a rocker. The exposure concentration was measured as the highest that caused no toxicity at 24 hours, as assayed by LDH release. For each exposure group, three wells were treated with Trizol at 2 and 24 hours after exposure. Then, all of these samples were performed RNA-seq to analyze whole-genome transcriptome using Illumina HiSeq 2500 platform with 50 bp single-end reads (Illumina, USA).
