Summary |
YTN2 and YTN16 were chemically induced gastric cancer cell lines. When inoculated subcutaneously, YTN2 spontaneously regresses while YTN16 grows progressively. By single-cell RNA-Seq and flow cytometry, we found more IL-17 producing cells in YTN16 than YTN2. Blockade of IL-17 signaling in YTN16 tumor-bearing mice suppressed tumor growth. To investigate tumor-infiltrating cells after IL-17 blockade, we performed single-cell RNA-Seq of CD45+ cells in YTN16 tumors.

Overall design |
Mice were inoculated with 5 ×106 YTN16 cells on day 0. Anti-IL-17 antibody (clone 17F3) was injected on days 0, 3, 6, and 9. On day 10, CD45+ cells were sorted from tumor tissues and processed by 10x controller with Single Cell 3' Reagent Kits v2.
