Summary |
ILC2s from different tissues manifest distinct properties. To investigate the molecular mechanisms regulating tissue features of ILC2s, we performed single cell RNA-seq on purified ILC2s from 5 different tissues.

Overall design |
ILC2s from the bone marrow, large intestine, lung, pancreas and small intestine from 6-week-old male wild-type mice were sorted by flow cytometry. ILC2s from bone marrow, large intestine, lung and pancreas were sorted as Lin(lineage)–ST2+CD25+ cells. And ILC2s from small intestine were sorted as Lin–Thy1intermediateCD45+KLRG1+ cells. Lineage markers were CD3, B220, CD11b, CD11c and FceRI. 30000 ILC2s from 5 tissues of 10 mice were subjected to scRNA-seq and two rounds of sequencing were performed on the each library of ILC2s from every tissue to obtain abundant reads and pooled for subsequent analysis.
