Summary |
We have shown previously that myocardial-associated B cells are transcriptionally distinct from circulating B cells. To investigate whether the sub-populations of B cells identified in the heart might not be unique to the heart, we investigated the transcriptional profile of B cells isolated from blood, heart, lung, and liver.

Overall design |
We performed single cell sequencing of FACS-sorted B cells isolated from blood, and perfused heart, liver, and lung. Samples were collected from the same mice, at the same time, and underwent the same digestion and staining protocols. To identify the tissue origin of the B cells, samples were stained with Hashtag antibodies, processed within the same 10X cDNA library and sequenced together to eliminate potential “batch” artifacts. We used the Chromium Single Cell 5′ Chromium instrument (from 10x Genomics) to perform single cell transcriptional profiling analyses.
