Title |
Tissue microenvironment shapes distinct sets of machineries to regulate ILC2 properties [ATAC-seq] |
Organism |
Mus musculus |
Experiment type |
Genome binding/occupancy profiling by high throughput sequencing
|
Summary |
ILC2s from different tissues manifest distinct properties. To investigate the molecular mechanisms regulating tissue features of ILC2s, we performed ATAC-seq on purified ILC2s from 5 different tissues.

Overall design |
ILC2s from the bone marrow, large intestine, lung, pancreas and small intestine from 6-week-old male wild-type mice were sorted by flow cytometry. ILC2s from bone marrow were sorted as Lin(lineage)–ST2+CD25+ cells. ILC2s for large intestine, lung and pancreas were sorted as Lin–CD45+CD127+ST2+CD25+. And ILC2s from small intestine were sorted as Lin–CD127+Thy1intermediateCD45+KLRG1+ cells. Lineage markers were CD3, B220, CD11b, CD11c and FceRI. Biological duplicates of 50000 ILC2s from 5 tissues of 10-16 mice were subjected to ATAC-seq sequencing analysis.
