Summary |
Meningeal immunity is critical to appropriate CNS function. Here we perform single cell RNA-sequencing to investigate how brain and meningeal stromal populations regulate immune trafficking and retention in young and old mice.

Overall design |
15 young (2 months) and 15 old (24 months) mice were sacrificed with euthasol and brain and dural meninges were collected. Tissues were dissociated with collagenase 8 to obtain a single cell suspension and sorted by FACS based on the following phenotypes: endothelial DAPI-Singlets CD45-CD31+CD13- and mural DAPI-Singlets CD45-CD31-CD13+. Equal amounts of endothelial and mural populations were pooled for single cell sequencing.
