Summary Meningeal immunity is critical to appropriate CNS function. Here we perform single cell RNA-sequencing to investigate how brain and meningeal stromal populations regulate immune trafficking and retention in young and old mice.
 
Overall design 15 young (2 months) and 15 old (24 months) mice were sacrificed with euthasol and brain and dural meninges were collected. Tissues were dissociated with collagenase 8 to obtain a single cell suspension and sorted by FACS based on the following phenotypes: endothelial DAPI-Singlets CD45-CD31+CD13- and mural DAPI-Singlets CD45-CD31-CD13+. Equal amounts of  endothelial and mural populations were pooled for single cell sequencing.
