Summary |
Gene expression changes specific for healthy undifferentiated and differentiated human gastric primary epithelial cells of different regions of the stomach were here determined in order to describe the transcriptomic profile of these cells in correlation to their DNA methylation pattern. In addition, gene expression changes upon 5-aza-2'-deoxycytidine treatment of antral gastric primary cells compared to mock-treated cells were deterimend.

Overall design |
Primary gastric epithelial cells were isolated from gastric sleeve resection material and cultivated as musosoids for 1-2 passages. RNA was isolated from the filter following the Trizol extraction method and gene expression was determined by Agilent 8x60k human custom single-color arrays. Three biological replicates of six different conditions (undifferentiated and differentiated antrum, corpus, and fundus samples) were analyzed as healthy gastric primary cells and two biological replicates of 5-aza-2'-deoxycytidine-treated and mock-treated undifferentiated antral samples were analyzed.
