Summary |
Calpain-mediated cleavage transfroms the full-length JP2 from a structural protein into a transcriptional regulator (JP2NT). We used RNA-seq to reveal the effect of JP2NT overexpression on cardiac transcriptional reprogramming induced by left ventricular pressure overload using transverse aortic banding in mice. We used ChIP-seq to reveal the genomic binding pattern of JP2NT. Additional ChIP-seq analyses against TBP and MEF2C were performed to reveal whether JP2NT overexpression affects endogeneous binding of TBP and MEF2C.

Overall design |
Cardiac specific overexpression of HA-tagged JP2NT transgene was achieved by an a-MHC-promoter driven transgenic system. The JP2NT-OE and littermate controls were subjected to transverse aortic banding (TAB) for 3 weeks. Left ventricle RNA was subjected to RNA-seq. Baseline JP2NT-OE or control hearts were subjected to ChIP-seq using antibodies against HA tag, MEF2C or TBP.
