Overall design |
C57BL/6J mice were fed High Fat Diet (Research Diet, D12331) and control diet (Research Diet, D12329) for 12 weeks. Mice were sacrificed by cervical dislocation every 4 hours along the day, liver collected and snap frozen. ChIP was performed as previously described (Uhlenhaut et al. 2012). Sheared chromatin was immunoprecipitated with anti-GR antibody (Santa Cruz, sc-1004X), anti-PPARalpha antibody (Santa Cruz, sc-9000), and a mixture of anti-Stat5a/b (RD System, AF2168) and anti-Stat5a/b (pY694/Y699) (Cell Signaling, 9351). Two replicates per condition and time point were used, for a total of 24 GR (ZT0-ZT20, control/HFD), 4 PPAR alpha (ZT12, control/HFD), and 4 STAT5 (ZT12, control/HFD) ChIP-seq samples.
