Summary
We identified a histone H3.3 sub-variant, H3mm7, to be specifically expressed in skeletal muscle satellite cells. H3mm7 knockout mice demonstrated an essential role of H3mm7 in skeletal muscle regeneration. Chromatin analysis revealed that H3mm7 facilitates transcription by forming an open chromatin structure around promoter regions including those of myogenic genes.
Overall design
We performed CEL-Seq2 in mouse tissues, RNA-seq and ATAC-seq in C2C12 and NIH3T3 cells, ChIP-seq in NIH3T3 cells. (BioProject: PRJDB5940, Accession: DRA005975, DRA006453)
