Manufacture protocol 1.2 nl of 10 µM HPLC purified, 5’ amino-modified DNA oligonucleotides (Metabion) were spotted four times on aminopropyltrimethoxysilane / phenylendiisothiocyanate coated glass slides using a customized piezoelectric spotter.
 
Description Each microarray contained a total of 256 DNA oligonucleotides with a sequence being reverse complement to the respective mature miRNA or control RNA, representing a total of 22 miRNA sequences (standard), 29 different variants of oligonucleoties and 13 controls. All probes were spotted in quadruplicates. The 22 miRNA sequences covered sequence verified miRNAs as deposited in the miRBase sequence database version 10.0. The 29 different variant oligonucleotides were chosen as follows: 9 different oligonucleotide variants for miR-122a and miR-16, including probes with (a) spacer sequences (6, 12 or 18 nucleotides in length named SP6, SP12 and SP18); (b) variants with omitted linker named NO-LINK; (c) dimer (DI) and trimer (TRI) variants and (d) oligo variants with adjusted TM (MELT). Oligo variants with adjusted TM were added for seven further miRNAs besides miR-16 and miR-122a. The 13 controls presented 10 positive and 3 negative controls.
