  1. The Alg44 alginate copolymerase constituted a stable dimer, with its periplasmic domains required for protein localization and alginate polymerization and modification.

  2. Alg44 may act as a co-polymerase in concert with Alg8, the major GT, and that both inner-membrane proteins are required in vivo for the polymerization reaction leading to alginate production

  3. Alg44 is a single pass transmembrane protein containing a cytoplasmic PilZ domain that binds the second messenger c-di-GMP in vitro. Point mutations abolishing binding in vitro also abolish function in vivo. Alg44 is required for alginate biosynthesis.
