Analysis of the accumulation of mutations in self-replicating RNAs using barcoded EGFP library

We engineered an orthogonal alphaviral RNA replication system to evolve synthetic RNA-based devices, enabling RNA replicase-assisted continuous evolution (REPLACE) in live mammalian cells.
