Regulation of c-Myc function by centromeric protein KLIP1

(Submitter supplied) In this study, we show that the centromeric Kaposi’s sarcoma-associated herpesvirus (KSHV) latent nuclear antigen (LANA)-interacting protein 1 (KLIP1) is a novel regulator of the c-Myc pathway. A series of microarray experiments were designed to demonstrate that the expression levels of KLIP1 protein are inversely correlated with cell proliferation, and the expression of c-Myc targets.

Glutaminyl cyclase (QC) activity affects gene expression in macrophages during inflammation

(Submitter supplied) Glutaminyl cyclase (QC) activity in macrophage cells is correlated with the gene expression of MCP-2 and QC-catalyzed N-terminal pGlu formation of MCPs is required for macrophage migration and provide new insights into the role of QC in the inflammation process.

Glycyrrhizin, silymarin, and ursodeoxycholic acid regulate a common hepatoprotective pathway in HepG2 cells

(Submitter supplied) Background: Glycyrrhizin, silymarin, and ursodeoxycholic acid are widely used hepatoprotectants for the treatment of liver disorders, such as hepatitis C virus infection, primary biliary cirrhosis, and hepatocellular carcinoma. Hypothesis/Purpose: However, few studies have evaluated the effects of these compounds on gene expression. Methods: Here, we used an oligonucleotide microarray approach to study the gene expression profile in human hepatoma cells treated with 25 μM of hepatoprotective agents. more...

Bone Morphogenetic Protein Receptor Antagonists Inhibit Growth and Survival of Lung Cancer Cells Expressing Stem Cell Markers Oct4 or Nestin

(Submitter supplied) H1299 cells were stably transfected with the Oct4 promoter/GFP or Nestin promoter/GFP reporter vectors. By FACS, 106 cells expressing high levels of GFP were isolated and placed into cell culture for twenty-four hours. Total RNA was used. Bone morphogenetic proteins (BMP) are aberrantly expressed in most lung carcinomas. BMPs mediate cell fate decisions and self-renewal of stem cells. Inhibition of BMP signaling decreases the growth and induces cell death of lung cancer cells lines. more...

Effect of erythropoietin on primed leukocyte expression profile

(Submitter supplied) Resistance to erythropoietin (EPO) treatment is observed in a considerable proportion of anemic patients with chronic kidney disease. Previous reports suggest that inflammation is one of the major independent predictors of this resistance, and pro-inflammatory cytokines have been shown to inhibit erythropoiesis. The aim of this study was to investigate EPO-induced modification to gene expression in primary cultured leukocytes. more...

Effects on expression profile of acute congestive heart failure (ACHF) patients monocytes by treatment with conventional drugs.

(Submitter supplied) Several studies have shown the importance of immune and inflammatory mediators in the pathogenesis of heart failure. In clinical practice has been observed that many conventional drugs can modulate circulating levels of these mediators. Despite, there is poor understanding of the precise mechanisms of these drugs in regulating immune and inflammatory systems. Blood monocytes were isolated from 6 hospitalized patients in Intensive Cardiology Care Unit (ICCU) with symptomatic acute congestive heart failure (ACHF) (NYHA Class III-IV) before and after treatment with conventional drugs (ARBs, ACEIs, diuretics, and beta-blockers). more...

Genes array from human mesenchymal stem cells (hMSCs) with knockdown EZH2

(Submitter supplied) EZH2 plays an important role in stem cell renewal and maintenance by inducing gene silencing via its histone methyltransferase activity. EZH2 downregulation markedly enhances neuron differentiation of human mesenchymal stem cells (hMSCs)chromatin at promoters of EZH2 target genes.

Extremely low frequency electromagnetic fields (ELF-EMFs) effect on the genetic expression profile of a human keratinocyte line (HaCaT)

(Submitter supplied) Clinical evidence has shown that electromagnetic fields produce a benefical therapeutic result for wounds, however little is still known about their exact mechanism of action. Moreover, clinical results concerning skin tissue restoration are still debated. In the present study, we carried out gene expression profiling of a human keratinocyte line (HaCaT) submitted to 1 hour of ELF-EMF (frequency 50Hz, intensity 1 mT) in order to identify up- and downregulated genes by this stimulus, and to verify the presence of specific molecular pathways activation. more...

Cumulus cells genetic profile of patients stimulated with r-hLH or hCG evaluated with MicroArray analysis versus r-hFSH stimulated COCs

(Submitter supplied) The aim of this study was to evaluate if different ovaric stimulation protocols modify gene expression profiles in COCs of infertile women. mRNA pools in COCs are essential for correct meiosis and activation of oocyte during fecondation and their alterations lead to reduced differentiation and oocyte functionality. In particular we tested the action of 2 different stimulation protocol on COCs transcriptome by MicroArray analysis of COCs after stimulation with r-hLH or h-CG versus COCs stimulated with r-hFSH. more...

Gene expression profile of Wharton-jelly mesenchymal stem cells at 12th in vitro expansion cycle compared to Wharton-jelly mesenchymal stem cells at 4th in vitro expansion cycle

(Submitter supplied) Mesenchymal stem cells (MSCs) are generally considered the main tool box for cell-based therapies and compared to embryonic stem cells (ESCs), MSCs exhibit many advantages. It has been shown that UC matrix (the Wharton’s Jelly surrounding umbilical vessels) contains a great number of mesenchymal cells. The abundant amount of Wharton’s Jelly (WJ) makes it an attractive source of MSCs for cell-based therapies. more...

Whole genome gene expression profiles of normo- and dyslipidemic men after fish oil supplementation

(Submitter supplied) Beneficial effects of long-chain omega-3 polyunsaturated fatty acids (n-3 FAs) are generally well-known from epidemiological studies, but the various mechanisms of action are not completely clarified. Regulation of gene expression is one known mechanism of action, but only very limited data of regulated pathways in humans after n-3 FA supplementation are available. Up to now, no studies compared gene expression changes after n-3 FA supplementation between normolipidemic and dyslipidemic subjects. more...

Effect of c-MET inhibitor SU11274 upon sensitive (RPMI8226) or multi-resistant (RPMI8226/R5) Multiple Myeloma cell lines

(Submitter supplied) The c-MET signaling axis is increasingly implicated in tumorigenesis and chemioresistance. In this study, we investigated gene expression modifications induced by SU11274 (Selleck Chemicals, Boston, USA), a novel selective c-MET inhibitor, in a pair of isogenic multiple myeloma (MM) cell lines either sensitive (RPMI8226) or multi-resistant, highly c-MET expressing (RPMI8226/R5) cells. On the whole, RPMI8226/R5 cells after SU11274 were characterised by wider and diverse gene expression modifications than RPMI8226, indicating that c-MET over-expression, and its inhibition, is an important aspect of the adaptive response associated to drug resistance.

Early-stage breast cancer

(Submitter supplied) Copy number alteration profiling of human early-stage (stage I and II) breast cancers. We aimed to describe the commonly occurred chromosome alterations in early-stage breast cancer (stage I and II) and to explore the implications of the recurrently altered regions (RAR) on the patient prognosis. For this purpose, we analyzed the DNA copy number alterations (CNAs) across the whole genome using oligoarray-comparative genomic hybridization (CGH) in the discovery set of EBC patients.
