Uncertain significance for Generalized epilepsy-paroxysmal dyskinesia syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001161352.2(KCNMA1):c.59G>A (p.Gly20Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 20 of the KCNMA1 protein (p.Gly20Asp). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This missense change has been observed in individual(s) with autosomal dominant KCNMA1-related disorders (PMID: 34224328). In at least one individual the variant was observed to be de novo.
