NM_001374736.1(DST):c.17310C>A (p.His5770Gln) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H3651Q variant (also known as c.10953C>A), located in coding exon 60 of the DST gene, results from a C to A substitution at nucleotide position 10953. The histidine at codon 3651 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and glutamine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
