Uncertain significance for Fanconi anemia — the classification assigned by Sema4, Sema4 to NM_004629.2(FANCG):c.422G>A (p.Arg141His), citing Sema4 Curation Guidelines. This variant lies in the FANCG gene (transcript NM_004629.2) at coding-DNA position 422, where G is replaced by A; at the protein level this means replaces arginine at residue 141 with histidine — a missense variant. Submitter rationale: The FANCG c.422G>A (p.R141H) variant has not been reported in the literature to our knowledge. It was observed in 1/10356 chromosomes of the Ashkenazi Jewish subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654) and has been reported in ClinVar (Variation ID 999134). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
