This variant lies in the ASS1 gene (transcript NM_054012.4) at coding-DNA position 247, where T is replaced by C; at the protein level this means replaces tyrosine at residue 83 with histidine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with histidine at codon 83 of the ASS1 protein (p.Tyr83His). The tyrosine residue is highly conserved and there is a moderate physicochemical difference between tyrosine and histidine. This variant is present in population databases (rs753659761, ExAC 0.002%). This missense change has been observed in individual(s) with citrullinemia (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant.
