NM_003995.4(NPR2):c.263A>C (p.Lys88Thr) was classified as Uncertain significance for Tall stature-scoliosis-macrodactyly of the great toes syndrome; Acromesomelic dysplasia 1, Maroteaux type by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 998732). This missense change has been observed in individual(s) with clinical features of acromesomelic dysplasia (PMID: 26284228; Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces lysine, which is basic and polar, with threonine, which is neutral and polar, at codon 88 of the NPR2 protein (p.Lys88Thr).
