NM_001903.5(CTNNA1):c.1753C>T (p.Pro585Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P585S variant (also known as c.1753C>T), located in coding exon 12 of the CTNNA1 gene, results from a C to T substitution at nucleotide position 1753. The proline at codon 585 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
