Uncertain significance — the classification assigned by Ambry Genetics to NM_002047.4(GARS1):c.1657A>G (p.Lys553Glu), citing Ambry Variant Classification Scheme 2023: The p.K553E variant (also known as c.1657A>G), located in coding exon 13 of the GARS gene, results from an A to G substitution at nucleotide position 1657. The lysine at codon 553 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
