NM_004174.4(SLC9A3):c.1421G>A (p.Arg474Gln) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 474 of the SLC9A3 protein (p.Arg474Gln). This variant is present in population databases (rs13154302, gnomAD 0.09%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with SLC9A3-related conditions. ClinVar contains an entry for this variant (Variation ID: 998416). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SLC9A3 protein function. Experimental studies have shown that this missense change affects SLC9A3 function (PMID: 25715704). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
