Submitter rationale: Variant summary: POU1F1 c.212C>T (p.Ala71Val) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.2e-05 in 250550 control chromosomes. c.212C>T has been reported in the literature in individuals affected with Combined Pituitary Hormone Deficiency (Fan_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
