This variant lies in the PKD1 gene (transcript NM_001009944.3) at 6 bases into the intron immediately before coding-DNA position 288 through 5 bases into the intron immediately before coding-DNA position 288, deleting this region. Submitter rationale: The PKD1 c.288-6_288-5del variant was not identified in the literature nor was it identified in the following databases: dbSNP, ClinVar, COGR, LOVD 3.0, ADPKD Mutation Database, PKD1-LOVD, the 1000 Genomes Project, the NHLBI GO Exome Sequencing Project, the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The c.288-6_288-5del variant is located in the 3' splice region but does not affect the invariant -1 and -2 positions. However, positions -3 and -5 to -12 are part of the splicing consensus sequence and variants involving these positions sometimes affect splicing. In addition, 3 of 4 in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) predict a greater than 10% difference in splicing.
