This variant lies in the PKD2 gene (transcript NM_000297.4) at 14 bases into the intron immediately before coding-DNA position 2020, where T is replaced by C. Submitter rationale: The PKD2 c.2020-14T>C variant was not identified in the literature nor was it identified in the dbSNP, ClinVar, GeneInsight-COGR, LOVD 3.0, ADPKD Mutation Database, or PKD1-LOVD, databases. The variant was not identified in the following control databases: the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The variant was identified by our laboratory in a patient with ADPKD and a co-occurring pathogenic PKD1 variant (c.11712+2T>C), increasing the likelihood that the c.2020-14T>C variant does not have clinical significance. The variant occurs outside of the splicing consensus sequence and in silico or computational prediction software programs (SpliceSiteFinder, MaxEntScan, NNSPLICE, GeneSplicer) do not predict a difference in splicing. In summary, based on the above information the clinical significance of this variant cannot be determined with certainty at this time although we would lean towards a more benign role for this variant. This variant is classified as likely benign.
