This variant lies in the BEST1 gene (transcript NM_004183.4) at coding-DNA position 272, where C is replaced by T; at the protein level this means replaces threonine at residue 91 with isoleucine — a missense variant. Submitter rationale: The T91I missense variant in the BEST1 gene has been reported previously in association with Bestdisease (Lotery et al., 2000; Querques et al., 2009). The T91I variant was not observed in approximately 6,500 individuals of European and AfricanAmerican ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variantin these populations. Therefore, we interpret R91I as a pathogenic variant.
