NM_000088.4(COL1A1):c.64G>C (p.Gly22Arg) was classified as Pathogenic for Osteogenesis imperfecta type I by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects COL1A1 function (PMID: 29101475). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 996641). This missense change has been observed in individual(s) with osteogenesis imperfecta (PMID: 16786509, 30450527). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 22 of the COL1A1 protein (p.Gly22Arg).
