This variant lies in the PLOD1 gene (transcript NM_000302.4) at 3403 bases into the intron immediately before coding-DNA position 77, where T is replaced by C. Submitter rationale: The PLOD1 c.128T>C; p.Leu43Pro variant in the alternate transcript NM_001316320.1, also known as c.77-3403T>C in transcript NM_000302.3, is not reported in the medical literature or gene-specific databases, to our knowledge. This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The leucine at codon 43 is weakly conserved but computational analyses (SIFT: damaging, PolyPhen-2: benign) predict conflicting effects of this variant on protein structure/function. However, this variant occurs in a weakly conserved exon that is expressed at low levels in the Genotype-Tissue Expression (GTEx) database (see link), suggesting it may not occur in physiologically relevant transcript.
