NM_000558.5(HBA1):c.416C>G (p.Ser139Cys) was classified as Uncertain significance by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories, citing ARUP Molecular Germline Variant Investigation Process: The Hb Ecuador (HBA1: c.416C>G; p.Ser139Cys, also known as Ser138Cys when numbered from the mature protein, rs63749989) has been reported in the heterozygous state in at least one individual with no associated clinical symptoms (see link to HbVar). This variant is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The serine at codon 139 is highly conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Due to limited information, the clinical significance of the Hb Ecuador variant is uncertain at this time.
