This variant lies in the ABCA4 gene (transcript NM_000350.3) at coding-DNA position 5644, where A is replaced by G; at the protein level this means replaces methionine at residue 1882 with valine — a missense variant. Submitter rationale: Variant summary: ABCA4 c.5644A>G (p.Met1882Val) results in a conservative amino acid change located in the ABC-2 type transporter, transmembrane domain (IPR013525) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251436 control chromosomes. c.5644A>G has been reported in the literature in compound heterozygous individuals affected with Retinitis Pigmentosa (Del Pozo-Valero_2020). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
