Pathogenic — the classification assigned by GeneDx to NM_000350.3(ABCA4):c.5161_5162del (p.Thr1721fs), citing GeneDx Variant Classification (06012015): The c.5161_5162delAC deletion in the ABCA4 gene has been reported previously in association with Stargardt disease (Maugeri et al., 1999; Papaioannou et al., 2000; Briggs et al., 2001). The deletion causes a frameshift starting with codon Threonine 1721, changes this amino acid to a Histidine residue and creates a premature Stop codon at position 65 of the new reading frame, denoted p.Thr1721HisfsX65. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Therefore, we interpret this variant as pathogenic.
