Submitter rationale: The c.425G>A (p.Ser142Asn) variant identified in the HUWE1 gene substitutes a conserved Serine for Asparagine at amino acid 142/4375 (coding exon 7/84). This variant is absent from gnomAD and ExAC, suggesting it is not a common benign variant in the populations represented in these databases. In silico algorithms do not agree on the functional consequence of this variant, as it is predicted both Neutral (Provean; score: -2.39) and Damaging (SIFT; score: 0.006) to the function of the canonical transcript. This variant is absent from ClinVar and to our current knowledge has not been reported in affected individuals in the literature. This variant was identified as a de novo variant in a proband submitted for clinical testing. It is reported here as Likely Pathogenic.
