This variant lies in the ALMS1 gene (transcript NM_001378454.1) at coding-DNA position 4454, where T is replaced by C; at the protein level this means replaces isoleucine at residue 1485 with threonine — a missense variant. Submitter rationale: Variant summary: ALMS1 c.4451T>C (p.Ile1484Thr) (also known as c.4457T>C, p.Ile1486Thr) results in a non-conservative amino acid change located in the Alstrom syndrome repeat (IPR040972) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 249292 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.4451T>C in individuals affected with Alstrom Syndrome With Dilated Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
