This variant lies in the CRB2 gene (transcript NM_173689.7) at coding-DNA position 952, where G is replaced by C; at the protein level this means replaces glycine at residue 318 with arginine — a missense variant. Submitter rationale: Variant summary: CRB2 c.952G>C (p.Gly318Arg) results in a non-conservative amino acid change located in the EGF-like domain (IPR000742) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3e-05 in 168030 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.952G>C in individuals affected with Focal Segmental Glomerulosclerosis 9 and no experimental evidence demonstrating its impact on protein function have been reported.
