NC_000017.10:g.(?_17140226)_(17140502_?)del was classified as Pathogenic for Birt-Hogg-Dube syndrome 1 by Division of Respiratory Medicine of Juntendo University, Juntendo University Faculty of Medicine and Graduate School of Medicine: This variant is a large genomic deletion including non-coding exon 1 of the FLCN gene. FLCN exon 1 functions as promoter for FLCN by luciferase reporter assay (PMID: 21412933). This variant has already been observed in several unrelated individuals affected with Birt-Hogg-Dube syndrome (PMID: 21412933). We demonstrated the genomic deletion including FLCN exon 1 caused a complete silencing of FLCN transcription from the affected allele of patients' cell. For these reasons, this variant has been classified as pathogenic.
