Pathogenic for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000249.3(MLH1):c.(207+1_208-1)_(545+1_546-1)del (p.Lys70Valfs), citing Invitae Variant Classification Sherloc (09022015): This sequence change is a gross deletion of the genomic region encompassing exons 3-6 of the MLH1 gene. This deletion extends to both edges of the assayed region, and, although the 5' and 3' boundaries of this event are in the 2nd and 6th introns, respectively, the exact breakpoints are not known. Gross deletions in MLH1 are known to be pathogenic. Deletions spanning exons 3-6 of the MLH1 gene have been reported in individuals affected with Lynch syndrome (PMID: 12938096, 16541406). For these reasons, this variant has been classified as Pathogenic.
