Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001034853.2(RPGR):c.106G>T (p.Val36Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 36 of the RPGR protein (p.Val36Phe). This variant is present in population databases (rs62638628, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with RPGR-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on RPGR function (PMID: 9990021, 23213406, 30622176).
