This variant lies in the EDAR gene (transcript NM_022336.4) at coding-DNA position 338, where G is replaced by A; at the protein level this means replaces cysteine at residue 113 with tyrosine — a missense variant. Submitter rationale: We detected the variant Cys113Tyr in EDAR in two members of a Chinese family and predicted its pathogenicity. SIFT, Polyphen2 and MutationTaster predictions for the mutation were â€œdeleteriousâ€ (0.00), â€œprobably damagingâ€ (0.996), and â€œdisease-causingâ€ (1.00), respectively, suggesting the variant was highly pathogenic. A cross-species amino acid sequence alignment of EDAR showed that the mutation site was highly conserved among humans (>NP_071731.1), cattle (>XP_005212787.1), zebrafish (>NP_001108536.2), rhesus monkeys (>XP_014968589.2), dogs (>XP_005626028.2), mice (>NP_034230.1), and chickens (>NP_001012629.1). In summary, the Cys113Tyr variant meets our criteria to be classified as pathogenic based upon segregation analysis and functional prediction results.
