Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_003242.6(TGFBR2):c.1013C>T (p.Thr338Met), citing ACMG Guidelines, 2015. This variant lies in the TGFBR2 gene (transcript NM_003242.6) at coding-DNA position 1013, where C is replaced by T; at the protein level this means replaces threonine at residue 338 with methionine — a missense variant. Submitter rationale: This missense variant replaces threonine with methionine at codon 338 of the TGFBR2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with Loeys-Dietz syndrome (PMID: 37090272) and in an individual affected with thoracic aortic aneurysm and dissection (PMID: 27879313). This variant has been identified in 1/31396 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
