Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016239.4(MYO15A):c.10373del (p.Leu3458fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the MYO15A gene (p.Leu3458Argfs*96). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 73 amino acid(s) of the MYO15A protein and extend the protein by 22 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MYO15A-related conditions. ClinVar contains an entry for this variant (Variation ID: 985835). This variant results in an extension of the MYO15A protein. Other variant(s) that result in a similarly extended protein product (p.Ser3525Alafs*29) have been determined to be pathogenic (PMID: 17851452). This suggests that these extensions are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
