Pathogenic for Retinitis pigmentosa 59 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_205861.3(DHDDS):c.104G>A (p.Gly35Glu), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DHDDS gene (transcript NM_205861.3) at coding-DNA position 104, where G is replaced by A; at the protein level this means replaces glycine at residue 35 with glutamic acid — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 35 of the DHDDS protein (p.Gly35Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of developmental and epileptic encephalopathy (PMID: 34382076; internal data). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 985622). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt DHDDS protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects DHDDS function (PMID: 34382076). For these reasons, this variant has been classified as Pathogenic.
