Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_198407.2(GHSR):c.820C>T (p.Leu274Phe), citing Ambry Variant Classification Scheme 2023: The alteration results in an amino acid change:_x000D_ _x000D_ The c.820C>T (p.L274F) alteration is located in coding exon 2 of the GHSR gene. This alteration results from a C to T substitution at nucleotide position 820, causing the leucine (L) at amino acid position 274 to be replaced by a phenylalanine (F). The alteration is rare in population databases: _x000D_ _x000D_ Based on data from the Genome Aggregation Database (gnomAD), the c.820C>T alteration was observed in 0.0021% (6/281586) of total alleles studied. The altered amino acid is conserved throughout evolution:_x000D_ _x000D_ The p.L274 amino acid is conserved in available vertebrate species. The alteration is predicted tolerated by in silico modeling:_x000D_ _x000D_ The p.L274F alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
