This variant lies in the RASA2 gene (transcript NM_006506.5) at coding-DNA position 1886, where G is replaced by A; at the protein level this means replaces arginine at residue 629 with glutamine — a missense variant. Submitter rationale: Variant summary: RASA2 c.1886G>A (p.Arg629Gln) results in a conservative amino acid change located in the Ras GTPase-activating domain (IPR001936) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250928 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1886G>A in individuals affected with Noonan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
