Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003242.6(TGFBR2):c.1168G>A (p.Asp390Asn), citing Ambry Variant Classification Scheme 2023: The p.D390N variant (also known as c.1168G>A), located in coding exon 4 of the TGFBR2 gene, results from a G to A substitution at nucleotide position 1168. The aspartic acid at codon 390 is replaced by asparagine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with thoracic aortic aneurysm or dissection (Weerakkody R et al. Genet Med, 2018 Nov;20:1414-1422; Li Y et al. Eur J Hum Genet, 2021 Jul;29:1129-1138). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
