Submitter rationale: The p.I1918T variant (also known as c.5753T>C), located in coding exon 39 of the NF1 gene, results from a T to C substitution at nucleotide position 5753. The isoleucine at codon 1918 is replaced by threonine, an amino acid with similar properties. This alteration was identified in a 2 year old with cafe au lait macules and freckling (Nemethova M et al. Ann Hum Genet, 2013 Sep;77:364-79). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
