A presumed nomenclature of c.(?_-227)_(*8555_?)del has been designated for the purposes of this classification. The variant was absent in 16120 control chromosomes (gnomAD, Structural Variants dataset). Deletion of the entire MECP2 gene has been reported in the literature in individuals affected with Rett Syndrome (e.g. Shi_2005, Philippe_2006, Quenard_2006). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as pathogenic.
