NC_000023.11:g.(31774193_31819974)_(31820084_31836717)del was classified as Pathogenic for Dystrophinopathies by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exon 50 in the DMD gene. A presumed nomenclature of c.(7200+1_7201-1)_(7309+1_7310-1)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a frameshift (predicted protein level effect : p.Arg2401Leufs*9) in the DMD gene, a known mechanism of disease. The variant was absent in 16117 control chromosomes (gnomAD, Structural Variants dataset). c.(7200+1_7201-1)_(7309+1_7310-1)del has been reported in the literature in individuals affected with Dystrophinopathies (e.g. Elhawary_2018, Skuk_2004, Zhang_2019). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One ClinVar submitter (evaluation after 2014) cite the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
