Benign for Orofacial cleft 6, susceptibility to — the classification assigned by Laboratorio de Biologia Experimental, Instituto Nacional de Pediatria, Mexico to NM_006147.4(IRF6):c.-4+33C>G, citing ACMG Guidelines, 2015: This variant was identified in a male subject diagnosed as non-syndromic cleft lip palate; with unilateral right cleft lip. No relative was found to be affected. The subject was included in a group of CL/P patients for genetic association study. The variant was consider Benign by its location and effect and also has been reported in healthy cohorts as gnomAD (45 alleles).

The individual is diagnosed as case of non-Syndromic cleft lip palate, evidence suggest that this variant is not responsible of a syndromic form of orofacial cleft. In the study 172 individuals diagnosed as non-syndromic cleft lip palate.No relatives of this patient were affected.
