Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002087.4(GRN):c.473G>A (p.Cys158Tyr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GRN c.473G>A (p.Cys158Tyr) results in a non-conservative amino acid change located in the Granulin domain (IPR000118) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 1.2e-05 in 251342 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.473G>A in individuals affected with Neuronal Ceroid-Lipofuscinosis (Batten Disease) and no experimental evidence demonstrating its impact on protein function have been reported, although the variant has been reported in unaffected controls in the literature (e.g., vanderZee_2007). The following publications have been ascertained in the context of this evaluation (PMID: 19020205, 17345602). ClinVar contains an entry for this variant (Variation ID: 98143). Based on the evidence outlined above, the variant was classified as uncertain significance.
