Likely pathogenic for Cornelia de Lange syndrome 3 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005445.4(SMC3):c.1417T>C (p.Trp473Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with arginine at codon 473 of the SMC3 protein (p.Trp473Arg). The tryptophan residue is highly conserved and there is a moderate physicochemical difference between tryptophan and arginine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with Cornelia de Lange syndrome (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 981413). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0").
